計劃內容: A preS2 mutant virus from patient can induce hepatocellular carcinoma
(H CC) in transgenic mice. The dysregulation of S gene expression is
involved in hepatocarcinogenesis. The purpose of this study is to
investigate molecular genetic and regulatory mechanisms whereby the
mutant induces HCC; transcriptional repressor, repressor complexes,
activators are involved in the deregulation of S gene expression.
Identification and functional characterization of transcription factors
involved in S gene expression are the main aim of this study.
工作內容: Recombinant DNA technologies (PCR, gene cloning, EMSA, ChIP, etc),
RNA analysis (RNAse protection assay, primer extension analysis), cell
culture (transfection), co-immunoprecipitation, immunoblotting,
bioinformatics, GST fusion protein, DNA affinity precipitation assay
(Dynabeads), yeast one hybrid system, etc.
