計劃內容: A hepatocellular carcinoma transgenic mice model, which is induced by
hepatitis B virus surface gene mutant (ie, preS2 mutant virus from a HCC
patient). The molecular genetic and regulatory mechanisms whereby the
mutant induces HCC are under investigation; transcriptional repressor
complexes, activator are involved in the dysregulation of S gene expression.
Identification and functional characterization of transcription factors involved
in S gene expression.
工作內容: Recombinant DNA technologies (PCR, gene cloning, EMSA, ChIP, etc),
RNA analysis (RNAse protection assay, primer extension analysis), cell
culture (transfection), co-immunoprecipitation, immunoblotting,
bioinformatics, GST fusion protein, DNA affinity precipitation assay
(Dynabeads), yeast one hybrid system, etc.
